NMN

Nicotinamide Mononucleotide

NMN is a naturally occurring compound found in trace amounts in foods including broccoli, avocado, and edamame. Supplemental doses are orders of magnitude higher than dietary intake and are required to meaningfully influence systemic NAD+ levels.

Resveratrol

Trans-Resveratrol, standardised

Resveratrol is a polyphenol found in red grapes and certain other plants. It is a known activator of SIRT1 — a NAD+-dependent sirtuin central to metabolic regulation and cellular stress response. The combination of an NAD+ precursor with a sirtuin activator is a logical pairing: NMN supports the availability of NAD+ for sirtuin function; resveratrol provides additional sirtuin-activating signalling.

The Science

What Is NAD+?


AD+ is a coenzyme found in every cell of the human body. It is essential to two fundamental biological processes: energy metabolism (as a key molecule in the mitochondrial electron transport chain) and DNA repair (as a substrate for PARP enzymes and sirtuins, which are proteins involved in cellular maintenance and longevity signalling).

The challenge is that NAD+ levels decline with age significantly. Research from the lab of David Sinclair at Harvard Medical School has shown that NAD+ levels in middle-aged mice are roughly half those of young mice. Human studies have confirmed similar age-related declines. This has generated substantial scientific and commercial interest in strategies to restore NAD+ levels.

Why Not Supplement NAD+ Directly?


NAD+ itself is not well absorbed when taken orally its molecular size prevents efficient transport across intestinal membranes. Instead, the most effective strategy is supplementing with precursor molecules that the body converts into NAD+ intracellularly. The two most researched precursors are NMN (nicotinamide mononucleotide) and NR (nicotinamide riboside).

NMN vs NR What Is the Difference?


Both NMN and NR are precursors in the NAD+ biosynthesis pathway. NR is one step earlier in the pathway than NMN, meaning it requires an additional conversion step inside the cell. NMN, being one step closer to NAD+, has in theory a more direct route. Animal studies have consistently shown both to be effective at raising NAD+ levels. Human data, while more limited, is growing. A 2021 study in Cell Reports Medicine confirmed that oral NMN supplementation in men over 65 raised blood NAD+ levels significantly versus placebo.

Energy Metabolism and Mitochondrial Function


NAD+ is essential to the function of the mitochondrial electron transport chain the process by which cells produce ATP, their primary energy currency. Declining NAD+ levels are associated with reduced mitochondrial efficiency. Animal studies from the Sinclair lab have shown that restoring NAD+ levels via NMN reversed several age-related declines in mitochondrial function in aged mice.

DNA Repair


PARP enzymes which are heavily involved in detecting and repairing DNA strand breaks are NAD+-dependent. As NAD+ levels decline, so does the efficiency of this repair process. This is one of the proposed mechanisms linking NAD+ decline to the accelerated accumulation of cellular damage seen in ageing.

Sirtuin Activation


Sirtuins are a family of proteins involved in regulating cellular metabolism, stress response, and longevity pathways. They are NAD+-dependent they cannot function without adequate NAD+ availability. Sirtuin activity has been linked to caloric restriction benefits, mitochondrial biogenesis, and inflammatory regulation. Supporting NAD+ levels is one of the primary strategies for maintaining sirtuin function as we age.

Cognitive Health


NAD+ plays a role in neuronal energy metabolism and in the clearance of damaged proteins within brain cells. Preclinical models have shown that NMN and NR supplementation can improve cognitive performance in aged animals, reduce markers of neuroinflammation, and support synaptic plasticity. Human trials specifically investigating cognitive outcomes are currently underway.

Important Context


Much of the most compelling NAD+ research has been conducted in animal models, primarily mice. Human trials are ongoing but the body of human evidence, while promising, is still developing.
